The indica japonica hybrid sterility can be restored by wide compatibility gene S5n, which becomes an important resource to the utilization of heterosis between rice subspecies indica and japonica. The S5n gene has been cloned.Obviously, molecular marker technology has become a fast and effective means to identify the gene. According the 136 bp deletion in the DNA sequence of S5n, a new functional marker InDel -S5n was developed based on the agarose gel electrophoresis and polyacrylamide gel electrophoresis, which amplified 224 bp (gene S5n) or 360 bp (gene S5i or S5j). To validate the InDel-S5n, 10 conventional rice varieties and an F2 population of 230 plants were used, the experimental results indicated that the design marker could be detected relatively well in an agarose gel and polyacrylamide gel, and could
accurately identify whether the test material comprising the gene and genotype (homozygous or heterozygous). Therefore, the InDel-S5n is practical to be used for S5n gene screening, molecular marker assisted selection breeding. |
