| Using the callus of Cephalotaxus mannii for suspension cultivation, the factors influencing the establishment
of C. mannii cell suspension culture system, including culture medium, Vitamin, sucrose concentration, pH, inoculum
amount, were investigated. The alkaloid content in cell suspension culture system of C. mannii was determined by HPLC.
The results showed that the optimal medium for the cell suspension culture was MS+35 g/L sucrose+10 mg/L VB1, the
inoculum size was 6% and initial pH was 5.5-7.0. It was useful for cell growth and harringtonine formatio to adjust pH to
6.0 every 5 days in the experiment. The cell growth basically accorded with the Scurve in one culture cycle, the cycle
was 30 days, the greatest products was obtained at the 25th day. The cell viability sharply increased at first and gradually
decreased in later. |
