In order to provide basis for establishing rapid propagation system of Epimedium koreanum Nakai,
this research discussed the influencing factors of explant disinfection and bud induction. Regarding the buds as
explants, with different sterilization time(2 min, 4 min, 2+2 min, 6 min), varieties of minimal mediums (MS, B5, WPM)
and different concentrations of plant growth regulators including 6-BA, IBA and GA3, the research systematically
studied the induction and propagation conditions for tiller buds. The suitable method for sterilizing bud was to
disinfect it with 75% ethanol for 30 s, and then treat it with 0.1% HgCl2 for (2+2) min, which could control the
survival rate above 87.5%. The optimal medium for bud induction was WPM+1.5 mg/L 6-BA+0.5 mg/L IBA+0.5 mg/
L GA3, with the induction rate of 81.67%. The propagation medium suitable for buds was WPM+1.5 mg/L 6-BA+0.5
mg/L IBA. The disinfection method suitable for tiller buds and the medium combination suitable for induction袁
propagation of adventitious buds were screened out, to lay the foundation for establishing the rapid cultivation system
for tiller buds of E. koreanum Nakai. |
