The single factor and orthogonal experiments were adopted to optimize the mixed fermentation conditions
of marine bacteria AiL3 and CⅢ-1. Then the growth-promoting effects of AiL3 and CⅢ-1 were determined and
compared in pepper seedlings when the two strains fermentated seperately or mixed. The results showed that the medium
formula was consisted of yeast extract 10 g/L,yeast powder 5 g/L,sucrose 8 g/L,manioc starch 10 g/L,CaCl2·2H2O 0.2
g/L,NaCl 0.2 g/L,FeSO4·7 H2O 0.1 g/L,MnSO4·7 H2O 0.5 g/L. And the fermentation conditions were as follows the
initial pH at 7.0,the liquid volume 230 mL/500mL,inoculum 4%,temperature 28℃,speed 180 r/min,fementation time
52 h. Under the optimized fermentation medium and conditons, the amount of living cells and spores of the two marine
bacteria mixed fermentation increased by 100 times compared with the minimal medium. The production of living cells
was 5.0×1010 CFU/mL,the spores was 4.5×1010 CFU/mL,the rate of spores was more than 90%. The apparent indexes
including plant height, stem diameter, fresh weight of pepper plants were promoted significantly compared to blank group
when irrigated the mixed fermentation broths in pot experiment. |
