Abstract: |
Taking tube seedlings of endangered plant Begonia fimbristipula as materials,the best way of in vitro
preservation of germplasm was studied. The results showed that conventional medium 6-BA and NAA combination
was not suitable for germplasm in vitro preservation of B. fimbristipula,the survival rate was only 5% after 90 d. PP333
inhibited the differentiation of tube seedlings sprout plexus,significantly promoted the formation of adventitious roots,
after 180 d,only the survival rate of 3.0 mg/L PP333 treatment reached 55%,variation of germplasm in restoring growth.
ABA(0.5-2.0 mg/L)inhibited the growth of tube seedlings,after 180 d,the test-tube seedling survival rate was
100%,the somaclone was better after recovery training. 1/2 MS culture medium had the best effect on B. fimbristipula,
the survival rate was as high as 85% after 240 d. After restoring growth,the tube seedlings multiplication and their
growth were superior to other processings. 1/2 MS culture medium can be used as the best training method for germplasm
in vitro preservation of B. fimbristipula. |
