Effects of cadmium on plant root growth by laser confocal microscopy technique |

Abstract: |
In order to detect the toxicity of heavy metal cadmium( Cd) on plant by a simple,straightforward and non-invasive approach,laser confocal microscopy technique was used to study the effects of different concentrations( 0,50,75,100,and 75μmol/L ) of Cd on Arabidopsis thaliana root growth. Results showed that root growth was inihibited by Cd treatments( ≥ 75 μmol/L). Using fluorescent probes specific for reactive oxygen species( ROS) and calcium( Ca),data revealed that Cd was able to increase the concentrations of ROS and Ca accumulated in roots. Furthermore,experiments with probes that lable dead cells and apoptotic cells showed that the viability of root cells decreased with the increase of Cd concentration. Finally,pre-treatment of Ca chelating agent EGTA could decrease ROS concnentration at root cells,while ROS scavenger showed no obvious effect on Ca accumulation. That suggested that upon Cd treatment on the seedlings,Ca accumulation would affect ROS increase in the root cells,that may contribute to the Cd adverse effect on root growth. These studies may provide a feasible approach by using laser confocal miscrocopy on evaluation of Cd effect on plant and also provide experimental data to futher reveal the mechanism how heavy metals affected plant growth and development. |
