Screening of lignin degradation strain MZ-9 and optimizationof enzyme production conditions in compost |

Abstract: |
This experiment use the potassium ferricyanide color,guaiacol color and aniline blue( Azure -
B) fading circle method,and liquid fermentation experiment,measured enzyme activitives of Laccase( Lac) and
Manganese Peroxidase( MnP) were as high as 9.53 U/mL,9.53 U/mL,lignin degradation rate was 20.32%,from the
compost. An active strain MZ-9 is screened as a strong lignin degradation strain. Physiological and biochemical tests
and 16S rDNA sequence analysis showed that strain MZ-9 belonged to Bacillus and had 99.2% similar to Bacillus
licheniformis. Enzyme production showed that when the pH value was 8.0,the culture temperature was 37℃,
the inoculation amount was 8% and the culture time was 6 d,the Lac and MnP activities of strain MZ-9 were the
highest,and the degradation rate of lignin was 20.51%. Therefore,strain MZ-9 is an excellent lignin degradation
strain,has a very good development and utilization value in the lignin degradation,and can provide a reliable strain
resource support for the subsequent industrial production. |
