Preparation of tilapia skin polypeptide and its application in scald repair |

Abstract: |
By using tilapia skin as raw material,the optimum conditions for enzymolysis of tilapia skin with
papain and alkaline protease were obtained through enzyme screening and orthogonal test of enzymolysis. The tilapia
skin polypeptide was prepared by stepwise composite enzymolysis of the two enzymes under the optimal enzymolysis
conditions,and the repair effect of tilapia skin polypeptide on skin scald wounds of New Zealand rabbits was
investigated. The results showed that the optimum enzymolysis conditions of papain and alkaline protease were
as follows: the pH,temperature,substrate concentration and enzyme dosage were 6.5 and 9.5,55℃ and 45℃,
30% and 35%,0.5% and 0.4%,and the corresponding protein recovery rates were 90.54(±1.72)% and 89.22(±2.14)%. The protein recovery rate of composite enzymolysis increased by 5.03% and 6.58% compared to those
of papain and alkaline protease under the optimal enzymolysis conditions,respectively. The results of scald healing
experiment showed that the wound healing rates of the experimental group and the positive control group reached
more than 95% on the 17th day,which were significantly different from that of the model control group( P < 0.05).
There was no significant difference in wound healing rate between the experimental group and the positive control
group,and the healing effects were similar. Histopathological results showed that the recovery of scald wounds in
experimental group was the best,indicating that tilapia skin polypeptide had a great repair effect on skin scald
wounds. |
