Induction culture of callus and bud in Pachysandra terminalis |

Abstract: |
In order to rapidly propagate elite varieties of Pachysandra terminalis and screen out optimal culture
conditions for callus and bud induction ,we studied the effects of different culture conditions on induction of callus
and bud in P. terminalis including different sampling time,disinfecting methods of explants,hormone concentration
andlight treatment,taking annual tender stem with lateral buds from P. terminalis as explants. The results showed that
the best sampling season was May which provided acallus induction and bud induction rate of explants of 44% . Better
disinfection effect could be achieved by disinfecting with 0.1% HgCl2 for 6 min,of which the pollution rate and the
death rate was 11% and 22% respectively. The optimal concentration of hormone for inducing medium of callus and
buds was 0.5 mg/L NAA+2.0 mg/L 6-BA,with an induction rate of 47%. Light culture is better than dark culture,
with an induction rate of 60%. |
