Cloning and expression analysis of transcription factor gene ZoNAC17 in ginger |

Abstract: |
NAC transcription factor plays an important role in the response of plant stress. To further explore
the structure and function of NAC transcription factors,based on the ginger transcriptional database,using RTPCR
method to clone the transcription factor gene ZoNAC17 of ginger,and analyze the ZoNAC17 expression pattern
of different tissues of ginger by digital expression profile. The sequence analysis results showed that the length of
ZoNAC17 gene’s ORF in ginger is 1 815 bp,encoding 604 amino acids residues,molecular weight is 67.52 kD.
ZoNAC17 transcription factor is a transmembrane protein without signal peptide,the isoelectric point is 4.59,and
the subcellular location is located in the nucleus. The phylogenetic analysis indicated that ZoNAC17 gene in ginger
had the closest genetic relationship with MaNAC17 gene of Musa acuminata. The results of digital expression profile
displayed that ZoNAC17 gene is strongly expressed in different tissues of ginger,and the expression level of ZoNAC17
in mature tissues is different,which is higher in stem than leaf,and is more in leaf than rhizome. During the
development of rhizomes of ginger,the expression level of ZoNAC17 increased first and then decreased gradually,
and the highest expression level is in the development period of about 3 months. |
