The shoot bases of Kaempferia elegans sterile seedlings were used as explants which were subcultured for propagation, induced for multiple shoots, and rooted to produce rooted plants, and their medium for tissue culture were screened to establish a rapid propagation system for K. elegans. The results showed that the appropriate medium for propagation of K. elegans were MS+6-BA 8~12 mg/L+NAA 0~0.1 mg/L+coconut milk 10%+sucrose 30 g/L+cara rubber powder 10 g/L, and the best propagation medium was MS+6-BA 8 mg/L+NAA 0.1 mg/L+coconut milk 10%+sucrose 30 g/L+cara rubber powder 10 g/L. The suitable medium for inducing multiple shoots were MS+6-BA 3~5 mg/L+NAA 0.05~0.1 mg/L+ coconut milk 10%+ sucrose 30 g/L+ cara rubber powder 10 g/L, and the most suitable medium for inducing multiple shoots was MS+6-BA 3 mg/L+NAA 0.05 mg/L+ coconut milk 10%+ sucrose 30 g/L+ cara rubber powder 10 g/L. The optimum rooting medium was MS+ NAA 0.1 mg/L+ mashed banana 10~15%+activated carbon 0.5~1.0 g/L+ sucrose 30 g/L+ cara rubber powder 9.5 g/L. |
