Evaluation of High and Low Oleic Acid Peanuts Based on Widely Targeted Lipidomics and Near Infrared Spectroscopy

Abstract:
      【Objective】A new detection method that can identify peanut varieties and evaluate the quality was explored, which could screen high oleic acid peanut-specific lipid biomarkers.【Method】By using ultra performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS), agronomic traits investigation, near infrared spectroscopy(NIRS)analysis and widely targeted lipidomics(WTL)research were conducted on two peanut varieties with significant differences in oleic acid content, namely, the high oleic acid variety KN1715 and the low oleic acid variety JH7H.【Result】The results of agronomic traits determination showed that, compared with JH7H, the length of the main stem and first lateral branch of KN1715 were significantly increased by 11.72% and 33.74% respectively; while the number of pods per plant, full pods rate per plant, hundred pod weigh, hundred kernel weight, and shelling rate were significantly decreased by 46.23%, 5.02%, 5.35%, 11.68% and 17.17%, respectively. Quality traits evaluation displayed that contents of oleic acid, stearic acid, arachidic acid, protein, amino acids, valine, methionine, isoleucine, leucine, phenylalanine, histidine, arginine and proline of KN1715 significantly higher than those of JH7H, but the contents of linoleic acid, palmitic acid, behenic acid and eicosanoic acid showed the opposite result. The WTL analysis exhibited that there were great differences in lipid metabolites between high and low oleic acid varieties, and a total of 16 classes of 295 lipid molecules were screened, and then 31 core differential lipid molecules in three classes were screened by multivariate statistical analysis of differential metabolites. Finally, the core differential lipid metabolites and quality indexes were combined with clustering and correlation analysis to obtain visualized phenotypic and lipid metabolite characteristics profiles.【Conclusion】The method can be well implemented to evaluate the variety and quality of peanut varieties at the level of phenotype and lipid molecules.
