【Objective】The contents of important active ingredients such as chlorogenic acid, rutin, and solanesol
in different tobacco varieties for propagating were detected by HPLC method with a view to providing references for the
effective development and utilization of tobacco germplasm resources.【Method】The contents of chlorogenic acid, rutin,
and solanesol in leaves of 17 tobacco germplasm resources for propagating at mature stage were determined by HPLC. The
chromatographic conditions were as follows: chromatograph was Agilent 1260, chromatographic column was VP-ODS,
flow rate was 1.0 mL/min, the column temperature was 35℃ and the detector was 1260 VWD.【Result】The content of
chlorogenic acid was the highest among the six isomers, and was detected in all 17 germplasm resources, and that of S16 Dabinliuyeyan sun-cured tobacco was the highest, followed by cryptochlorogenic acid and neochlorogenic acid. Compared
with the first three isomers, the content of isochlorogenic acid A, isochlorogenic acid B and isochlorogenic acid C was
generally lower. The content of rutin in NC347 flue-cured tobacco was the highest(46.456 g/kg), followed by S19 Dajianye
(7.782 g/kg)and S17 Daduqinggeng(6.250 g/kg)sun-cured tobaccos, and that of Yunyan 87 flue-cured tobacco was the
lowest(1.206 g/kg). The content of solanesol in NC347 flue-cured tobacco was the highest(44.913 g/kg), followed by
K326 flue-cured tobacco(11.140 g/kg)and S17 Daduqinggeng air-cured tobacco(9.844 g/kg), and that of Huangkeng
No.7-1 flue-cured tobacco was the lowest(2.793 g/kg).【Conclusion】Among the 17 tobacco germplasm resources, the
content of rutin and solanol was higher than that of chlorogenic acid, and the highest level was found in NC347 flue-cured
tobacco. The content of chlorogenic acid was the highest among the six isomers, among which the S16 Dabinliuyeyan suncured tobacco had the highest content. The tobacco germplasm resources for propagating did not topping after the appearance
of flower-buds, which may have a greater effect on chlorogenic acid. |
