      【目的】WRKY 转录因子家族在植物生长发育、抵御胁迫等过程起着至关重要的作用。挖掘参与冬瓜(Benincasa hispida Cogn.)非生物胁迫的 WRKY 基因家族成员,探究其生物学功能。【方法】基于冬瓜基因组数据库鉴定冬瓜 WRKY 成员,利用生物信息学的方法系统分析其 WRKY 基因家族成员的蛋白理化性质、系统发育、保守基序、顺式作用元件,通过 qPCR 分析冬瓜 WRKY 的表达情况。【结果】冬瓜 WRKY 基因家族有 57 个成员,氨基酸数目在 126~650 之间,相对分子质量在 13.92~71.68 kD 之间;蛋白等电点在 4.61~9.69 之间。根据系统进化树将该家族分为 3 个类群,第二类群又分为 5 个亚组。冬瓜 WRKY 外显子有 2~6 个。染色体定位分析发现,有 56个基因定位在 12 条染色体上,1 个基因未定位在染色体上。保守基序分析显示,Motif 1 和 Motif 3 存在于 56 个基因,且同一类群具有类似的保守基序结构。同时,冬瓜 WRKY 基因的启动子上均含有应激反应相关元件、激素响应元件。WRKY 在冬瓜不同组织和果实发育时期特异表达。RT-PCR 结果表明非生物胁迫和激素处理冬瓜可诱导部分 WRKY基因的表达。【结论】共鉴定出 57 个冬瓜 WRKY 基因,同亚组的成员具有类似的保守基序结构。冬瓜 WRKY 基因在不同组织中的表达存在差异,其中,BhWRKY2、BhWRKY5、BhWRKY39、BhWRKY11、BhWRKY31、BhWRKY53参与不同的胁迫响应。

      【Objective】WRKY transcription factor family plays an important role in plant growth, development and stress resistance. The biological functions of WRKY gene family members involved in abiotic stress of Benincasa hispida Cogn. (wax gourd) were explored.【Method】WRKY members of wax gourd were identified based on the genome database of wax gourd. The protein physicochemical properties, phylogeny, conserved motifs and cis-acting elements of the members of the BhWRKY gene family was systematically analyzed by bioinformatics method. And the expression of wax gourd WRKY was analyzed by qPCR.【Result】The results showed that there were 57 members in the BhWRKY gene family with amino acid numbers ranging from 126 to 650 and molecular weight ranging from 13.92 to 71.68 kD. The isoelectric point of protein ranged from 4.61 to 9.69. According to phylogenetic tree, the family was divided into 3 groups, and the second group was further divided into 5 subgroups. The number of WRKY exons of wax gourd ranged from 2 to 6. Chromosomal mapping analysis showed that 56 genes were located on 12 chromosomes, and one gene was not located on the chromosome. The conserved Motif 1 and Motif 3 were found in 56 genes, and the same group had similar conserved motif structure. Meanwhile, it was also found that the promoters of the BhWRKY genes all contained cis-responsive elements and hormone response elements related to stress response. WRKYs were specifically expressed in different tissues and fruit development stages of wax gourd. RT-PCR result showed that abiotic stress and hormone treatment could induce the expression of some BhWRKY genes.【Conclusion】A total of 57 WRKY genes of wax gourd are identified and members of the same subgroup have similar structures. There are differences in the expression of 57 WRKY genes in different tissues. Among them, BhWRKY2, BhWRKY5, BhWRKY39, BhWRKY11, BhWRKY31 and BhWRKY53 participate in different stress responses.
