      【目的】试验前期组学数据提示甲型流感病毒(Influenza A Virus,IAV)感染 A549 细胞后宿主的 环状 RNA(circular RNA,circRNA)hsa_circ_004389 表达显著上调。为进一步明确 hsa_circ_004389 表达和 IAV 感染间的关联,研究其对 IAV 复制的影响。【方法】(1)采用荧光定量 PCR 等方法,确定在 IAV 病毒感染后 不同时间点和不同感染复数(Multiplicity of Infection,MOI)条件下 hsa_circ_004389 的表达水平;(2)检测聚肌 苷酸胞苷酸(Polyinosinic acid:polycytidilic acid,poly I∶C)和细菌脂多糖(Lipopolysaccharide,LPS)处理后 hsa_ circ_004389 的表达情况;(3)干扰素(Interferon,IFN)β 和 JAK 的抑制剂处理细胞后检测 hsa_circ_004389 的 表达情况;hsa_circ_004389 过表达和敲低后,在 mRNA 和蛋白水平上检测该环状 RNA 对 IAV 复制的影响。【结 果】宿主细胞内 hsa_circ_004389 表达的增加与 IAV 感染相关,且与病毒复制呈明显正相关,而 JAK-STAT 信号 通路参与 hsa_circ_004389 的表达调控,并且 hsa_circ_004389 能够抑制 IAV 复制。【结论】hsa_circ_004389 是宿 主应答 IAV 感染的产物,为潜在的细胞Ⅰ型 IFN 应答 IAV 的拮抗因子。

      【Objective】Based on the preliminary transcriptome profiles of A549 cells responding to influenza A virus (IAV) infection, one of the most significantly up-regulated host circular RNA(circRNA), hsa_circ_004389, was selected to further validate its expression and explore its roles in IAV replication.【Method】First, real-time quantitative PCR (qPCR) was used to verify the expression levels of hsa_circ_ 004389 at different time courses and different multiplicity of infection (MOI) of IAV infection in A549 cells; Second, hsa_circ_ 004389 expression in A549 cells treated with poly: I∶C (polyinosinic acid ∶ polycytidylic acid) and LPS (Lipopolysaccharide) were detected, respectively; Third, A549 cells were treated with interferon beta (IFNβ) and JAK inhibitor, and the expression of hsa_circ_004389 was determined; Finally, after a transient overexpression and knockdown in A549 cells, IAV replication at mRNA and protein levels were determined.【Result】 There was a positive correlation between the expression of hsa_circ_004389 and IAV replication. JAK-STAT signaling path was involved in regulating hsa_circ_004389 expression, and hsa_circ_004389 had the ability to inhibit IAV replication. 【Conclusion】Hsa_circ_004389, the product of the host responding to IAV infection, is a potential host anti-IAV factor.
