【目的】生长素是一种必需的植物激素,而 AUX/IAA 是生长素原初响应因子在环境胁迫中植物
生长和发育中起着至关重要的作用。为了解 VaIAAs 在小豆生长发育过程中的生物学功能,对 VaIAAs 基因家族
进行全基因组鉴定和分析。【方法】利用生物信息学对小豆的 AUX/IAA 进行全基因组鉴定和分析,基于 RNAseq 分析 VaAUX/IAA 基因家族在各组织的表达模式。【结果】在小豆全基因组共鉴定到 41 个 VaIAAs 基因,亚
细胞定位显示均为定位为细胞核。系统发育分析显示来自小豆、拟南芥(Arabidopsis thaliana)和水稻(Oryza
sativa)的 AUX/IAA 蛋白聚集成 6 组。基因结构分析表明,所有基因均含有内含子数目 1~8 个,VaIAAs 基因包
含 1~21 个外显子。顺式作用元件分析发现参与响应生物 / 非生物胁迫顺式作用元件最多,包括最常识别到的
ATTATA.bos、G-box 和 sp1 基序,还发现富集在厌氧诱导响应元件 ARE 和植物抗逆相关的 MYB 元件。蛋白 -
蛋白互作网络分析发现,所有 VaIAAs 均与 ARF 相连,此外还有部分基因与 AXR3(生长素诱导阻遏因子)、MP
(转录因子 B3 家族蛋白)等基因互作。VaIAAs 具有组织特异性,在不同组织的基因表达水平差异较大。
【结论】小豆 AUX/IAA 基因家族的基因结构、系统进化、蛋白质 - 蛋白质网络互作等生物信息学分析结果,将
为揭示小豆 AUX/IAA 基因家族在小豆生长发育过程中的功能提供重要的线索。 |

【Objective】Auxin is an essential plant hormone, and AUX/IAA is an important auxin initial response
factor that plays a crucial role in the growth and development of plants under environmental stress. To understand the
biological function of VaIAAs in the growth and development of Vigna angularis, genome-wide of VaIAAs gene family
was identified and analyzed.【Method】Bioinformatics was used to identify and analyze genome-wide of AUX/IAA in
Vigna angularis. Expression patterns of VaAUX/IAA gene family in various tissues were analyzed based on RNA-seq.
【Result】A total of 41 VaIAAs genes were identified in genome-wide of V. angularis, and subcellular localization showed
that all of them were localized to the nucleus. Phylogenetic analysis revealed that AUX/IAA proteins from V. angularis,Arabidopsis thaliana, and Oryza sativa clustered into six groups. Gene structure analysis showed that all the genes contained 1-8
introns and VaAUX/IAA genes contained 1-21 exons. Analysis of cis-acting elements found that cis-acting elements involved
in response to biological/abiotic stress were the most, including the most commonly recognized ATTATA.bos, G-box and sp1
motifs. The enrichment of anaerobic-induced response element ARE and plant stress resistance-related MYB elements was
also found. Protein-protein interaction network analysis showed that all VaIAAs were linked to ARF, in addition, some genes
interacting with AXR3 (auxin-induced repressor), MP (transcription factor B3 family protein), and etc. VaIAAs were tissuespecific, with large differences in gene expression levels in different tissues.【Conclusion】The results of bioinformatics
analysis of gene structure, phylogeny, and protein-protein network interactions of the V. angularis AUX/IAA gene family will
provide important clues to reveal the functions of AUX/IAA gene family in the growth and development of V. angularis. |
